Research on the Mechanical Control of AMD Pathogenesis

Recent work from Professor Kaustabh Ghosh’s lab, which implicates endothelial cell (EC) stiffening as a crucial yet previously–unrecognized risk factor for dry age-related macular degeneration (AMD), is featured on the cover of the November 2016 issue of Investigative Ophthalmology and Visual Science (Vol. 57, Issue 14, Page 5910-5918).

This work, led by doctoral student Andrea Cabrera and Professor Ghosh, demonstrates that senescence associated with aging leads to stiffening of choroidal ECs that, in turn, increases their susceptibility to complement injury, a major risk factor for AMD. This complement-mediated injury of the stiffer senescent ECs may explain why choroidal vessels degenerate in aging eyes, a hallmark of dry AMD.

These novel findings form the basis for the BrightFocus Foundation Grant that the Ghosh lab received earlier this summer. As part of this funded research, carried out in collaboration with researchers at the Oregon Health Sciences University, the Ghosh lab is using a monkey model of AMD to evaluate the role of this novel mechanism of choroidal vascular degeneration in clinical AMD pathogenesis.
